Protease Production by Submerged Fermentation in Shake Flasks Using Bacillus sp. Isolated from the Soil

Proteases are one of the most industrially important enzymes, which account for about 60% total enzyme market. Protease production by submerged fermentation in shake flasks using Bacillus sp. isolated from soil was studied. Soil samples were collected different locations within Chukwuemeka Odumegwu Ojukwu University, Uli, Anambra state. The serially diluted and inoculated on sterilized skim milk agar plates. plates incubated at 30oC 72 h. A clear zone around colonies gave an indication protease-producing bacteria isolates. selected protease producers subsequently used flask 50 ml sterile medium. Optimization study conducted to determine effect carbon sources, nitrogen trace elements, agitation rates pH. Twenty isolates found be active RS-5 OS-9 had highest clearance 13.5 12.1 mm respectively. result revealed that accumulated maximum yield 3.23 2.71 U/ml Gram positive endospore formers, identified as OS-9.The addition Starch maltose stimulated optimum 3.47 2.77 Beef extract enhanced 3.35 2.90 Maximum 3.28 2.85 obtained supplementation 0.4 g/l FeS04 observed rate 200 rpm 170 OS-9. At pH8, accumulation harbours some strains can greatly through optimization process parameters.
